Title: Effects of Fufangdanshen on choline acetyl transferase and acetyl cholinesterase activities of hippocampus and cerebral cortex in rats with senile dementia
Abstract: Objective To investigate the effects of Fufangdanshen on choline acetyl transferase (ChAT) and acetyl cholinesterase (AChE) activities in hippocampus and cerebral cortex, and the spatial discrimination in rats with Altheimer's disease induced by amyloid beta peptide injection.Methods Aβ1~40 was injected into hippocampus of adult Sprague-Dawley rats, and all tests were performed 14 days later. The deposition of senile plaques and the formation of neurofibrillary tangles were observed by thioflavin-S stain.Morris water maze was used to monitor the spatial discrimination. Colorimetric method was used to detect the activity of ChAT and AchE.Results Neurofibrillary tangles and senile plaques were detected in the hippocampus in the model rats after Aβ injection. In spatial discrimination experiment, the rats showed a remarkable time prolonging in platform searching, with a significant increase in swimming distance after Aβ injection. The ChAT activity in both hippocampus and cerebral cortex decreased significantly after Aβ injection. There was no significant change observed in AChE activity though a little enhancement appeared. Administration of Fufangdanshen 1.05 g/kg and 0.525 g/kg significantly alleviated the impairment in spatial discrimination caused by Aβ;ChAT activity was clearly improved when Fufangdanshen was administrated after Aβ.We did not observe an obvious effect of Fufangdanshen on AchE activity.Conclusion Fufangdanshen could improve the impairment of spatial discrimination, and alleviate the brain ChAT activity decrease in AD rats induced by Aβ.
