Title: Protective effect of bromelain on myocardial ischemia-reperfusion injury in rats
Abstract: Objective To investigate the protective effects of bromelain (Br) on myocardial ischemia-reperfusion(I-R) injury.Methods Thirty-two SD rats were divided into 4 groupps with 8 rats each.The myocardial I-R rat model was established by occlusion of left arterial decending for 30 min and reperfusion for 2 hours in groups of B,D and C.Group A was sham operated.PBS preconditioning with PBS via intraperitoneal injection for 15 consecutive days before ischemia in group B,Br plus PBS was used in group C instead of PBS alone as in group B.Group D was treated with I-R only.The deviations of ST-segment on ECG were recorded during study.Serum creatine kinase brain isozyme(CK-MB) and lactate dehydrogenase(LDH) were measured at 2 hours during reperfusion.Myocardial infaction(MI) and risk area were determined by Evans blue and TTC staining.Myocardial cell apoptotic index(AI) was determined by TUNEL.The change of phospho-protein kinase B(P-AKT) was determined by Western blot.Results Serum CK-MB and LDH,MI size and AI were less,but P-AKT was higher,in group C than those in groups of B and D(P0.01).Conclusion Br preconditioning may protect the myocardium from I-R injury by increasing P-AKT.
