Title: Qualitative and Quantitative Study of the Flavonoids in Prunus pseudocerasus and Preliminary Evaluation of Its Antioxidant Activity in Vitro
Abstract: OBJECTIVE:To establish a qualitative and quantitative analysis method for the flavonoids in Prunus pseudocerasus,and to evaluate the antioxidant activity of the extract in vitro preliminarily. METHODS:After hydrolysis,total flavonoids were extracted. The contents of total flavonoids were determined by UV spectrophotometry. TLC was applied to detect quercetrin and kaempferol. The contents of quercetrin and kaempferol in the flavonoids were determined by HPLC. Fention method was applied to evaluate the antioxidant activity of the extracts in vitro. RESULTS:The linear range of rutin was 11.0~110.0 μg·mL-1(r=0.999 8) with an average recovery of 98.3%(RSD=1.3%,n=6) in the UV method;the quercetrin and kaempferol can be detected by TLC. the linear ranges of quercetrin and kaempferol were 37.5~1 200.0 ng·mL-1(r=0.999 7) and 62.5~2 000.0 ng·mL-1(r=0.999 4) . The average recoveries were 99.8%(RSD=3.4%,n=9) and 100.0%(RSD=1.9%,n=9) in HPLC method. The total flavonoids extracted from P. pseudocerasus showed antioxidant activity in dose-dependant manner. CONCLUSION:This method is accurate,reliable and reproducible. It can be used for the quality control of P. pseudocerasus.
