Title: Determination of Ethyl Glucuronide,A Quantification of Trace Metabolite of Alcohol in Blood Using Gas Chromatograpy-mass Spectrometry
Abstract: Objective To establish a gas chromatography(GC) with mass spectrometric(MS) detection method for the determination of ethyl glucuronide(EtG),a metabolite of alcohol in human blood. Methods After centrifugation of the 1 ml blood anticoagulated with EDTA-Na_2,the supernate was precipitated protein by perchloric acid,and then cleaned up by an NH_2-solid phase extraction cartridge.The extractions were treated with BSTFA +TMCS(99:1 v/v) for derivatization,and can be analyzed by gas chromatography-mass spectrometry in Scan and SIM mode simultaneously.The quantitation of EtG was done by using internal standard method. Results A good linearity was obtained within the concentration range of 0.2-4.0 mg /L,with the correlation coefficient of the standard curve was 0.998 8.The limit of detection(LOD) was 0.17 mg/L.The recoveries were between 75.3%-85.6%,and the relative standard deviation(RSD) were in 5.6%-7.6%. Conclusion This method is proved to be sensitive,specific,accurate,and can be applied to the determination of EtG for medicolegal identification and clinical laboratory.
