Title: Apoptosis of Human Colonic Cancer Cell Line Induced by Survivin Antisense Oligonucleotide
Abstract: Objective:To explore the apoptosis of human colonic cancer cell line LS174T induced by liposome-survivin antisense oligonucleotide.Methods: Survivin antisense oligonucleotide was synthesized and trasfected into LS174T cells by lipofection.Survivin expression was determined by reverse transcription-polymerase chain reaction(RT-PCR) and Western-blotting.The caspase-3 activities were measured.The apoptosis rate was detected by flow cytometry.Results: The mRNA expressions of survivin were inhibited by survivin ASODN(P0.05);we could not detect the expression of survivin protein by Western-blotting after treatment with survivin ASODN;The caspase-3 activity was significantly higher than that of control group;Apoptosis rate reached much higher than that of untreated group.Conclusion:Survivin antisense oligonucleotide can effectively induce apoptosis of human colonic cancer cell line LS174T.
