Title: A study on differentiation of rabbit's bone marrow mesenchymal stem cells into osteoblasts in vitro
Abstract: Objective To investigate the methods of culture of rabbit's bone marrow mesenchymal stem cells(MSCs)in vitro and the differentiation of MSCs into osteoblasts.Methods The MSCs were obtained by femoral trochanter aspiration from rabbit and purified by wall adhesion culture procedure and passaged in vitro.Passage 3 MSCs were chosen to induce into osteoblasts.Sixteen days later,Alkaline Phosphatase(AP)was detected with BCIP/NBT and calcium nodules were stained with chinalizarin.Results MSCs were obtained by wall adhesion culture procedure.The MSCs had good reproductive activity.AP's detection and calcium nodules' staining were strongly positive.Conclusion MSCs separated from rabbit's bone marrow can be induced into osteoblasts and used as seed cells in bone issue engineering.
