Title: Effects of IL-32γ on the proliferation and cell cycle of fibroblast-like synoviocytes in patients with rheumatic arthritis
Abstract: Objective:To explore the effects of IL-32γ on the proliferation and cell cycle of fibroblast-like synoviocytes(FLS) in patients with rheumatic arthritis(RA) and the possible molecular mechanisms.Methods:Fibroblast-like synoviosytes obtained from patients with active rheumatic arthritis and osteoarthritis(OA) were stimulated with IL-32γ.The proliferation of cell was examined by MTT.The cell cycles were analyzed by Flow Cytometry.The levels of cyclin D1 and NF-κBp65 protein were detected by Western blot.The expression of proliferating cell nuclear antigen(PCNA) was examined by immunocytochemical staining.Results:IL-32γ was not found to induce proliferation of cultured OA-FLS.Administration of 10-100 ng/ml IL-32γ for 72-120 h significantly promoted the proliferation of RA-FLS in a dose-and time-dependent manner.After stimulation with 100 ng/ml IL-32γ for 48 h,the cell cycle transition from G1 phase to S+G2 phase were accelerated and the expression levels of NF-κBp65 and cyclin D1 and PCNA increased as compared with those of control group.Conclusion:IL-32γ can promote RA-FLS proliferation and accelerate the cell cycle transition via upregulating the expression of NF-κBp65 and cyclin D1.
