Title: Effect of Transforming Growth Factor-β_1 Antisense Oligonucleotides on Renal Interstitial Cell
Abstract: 【Objective】To investigate the effect of transforming growth factor-β1(TGF-β1)on the proliferation and apoptosis of renal interstitial cell.【Methods】 HEK293 cell line was treated with TGF-β1 antisense oligodeoxynucleotides(ASODN) by lipofectamine.The inhibition of proliferation and apoptosis of the cell were studied with cell growth curves,MTT method and flow cytometry.The expression of TGF-β1 was detected by RT-PCR and Western blot respectively.【Results】After transfection,the mRNA and protein expression of TGF-β1 in the cells were significantly decreased or negative expression.Compared with the control group,TGF-β1 significantly inhibited cell proliferation and induced cell apoptosis(P0.01).【Conclusion】 TGF-β1 ASODN may significantly inhibit the proliferation and induce apoptosis of renal interstitial cell.These results suggest that infected TGF-β1 ASODN by lipofectamine has an important role in the treatment of renal fibrosis.
