Title: Sporulation conditions of Cylindrocarpon destructans
Abstract: In order to find out the conditions for sporulation of Cylindrocarpon destructans,the investigation of sporulation of C.destructans under different culture conditions was conducted.The results showed that the suitable temperature for sporulation of conidia ranged from 15 ℃ to 25 ℃ whereas the suitable temperature for sporulation of chlamydospores ranged from 10 ℃ to 20 ℃ with the optimum temperature of 15 ℃.The suitable pH value for conidia and chlamydospores was varied from 5 to 7 with the optimum pH of 6.All the pathogens cultured on various carbon source medium could produce conidia and chlamydospores.Among the carbon sources,glucose and fructose were better for producing conidia.Nevertheless,no obvious influence of carbon sources on chlamydospores production was observed.The pathogens could produce a greater amount of conidia on carbamide medium,but did not produce conidia on nitrogen deficient medium.All the pathogens cultured on different nitrogen source medium could produce chlamydospores;among these,the pathogens cultured on the yeast extract medium produced the highest amount of chalmydospores.The greatest amount of conidia was produced in less time when the pathogens was cultured on ginseng root medium,whereas a greater amount of chlamydospores was produced in less time when the pathogens was cultured on PDA and basic medium.Light conditions had not evident influence on conidia production,but had an obvious inhibition effect on chlamydospores production.
