Title: Conditions of lipase production by endophytic Aspergillus niger and enzymatic properties of the crude enzyme
Abstract: A lipase-producing strain Aspergillus niger C2J6 was screened from Xinjiang wine grape. The conditions for enzyme production and enzymatic properties of C2J6 were studied. The results showed that the suitable conditions for enzyme production were as follows: lactose 1% as carbon source, peptone 1% as nitrogen source, initial pH 8.0, culture temperature 35℃, incubation time 72 h. Under the optimal condition, the lipase activity reached 18.75 U/ml. The enzyme was medium temperature amylase, and the optimal reaction temperature, pH of crude enzyme produced by the strain was 40℃ and 7.0, respectively. It had good thermal stability, 54.55% of the enzyme activity could be reserved after being kept at 50℃ for 1 h. Its activity was stable between pH 3.0-7.0, and it had some acid-resisting features. Mn2+could boost up enzyme activity, whereas Zn2+, Fe2+, Cu2+ would restrain its activity, and K+, Ca2+, Na+, Mg2+have little effect on its activity. When the enzyme was in sunflower oil and grain as substrate, the enzyme activity was 228% and 180%, respectively, which was significantly higher than the olive oil and smoke lampblack machine oil.
