Title: Analysis of the Internal Transcribed Spacer(ITS) Sequences in rDNA of 10 Strains of Fusarium spp.
Abstract: [Objective] The aim of the research was to explore the phylogenetic correlations among the strains of Fusarium spp.[Method] The genomic DNAs was extracted from 10 isolated strains of Fusarium spp.by using benzyl chloride method and PCR amplification and sequencing were made on its internal transcribed spacer(ITS) sequence.The sequencing results were searched for the homology on GenBank website by using Blast method and the ITS phylogenetic tree with the related strains was constructed by neighbor-joining method.[Result] The tested strains were located in 3 clades of the phylogenetic tree. Seven strains and Fusarium solani were classified into one clade,with the sequence similarity of 98.61%~100%.Strain F94 and Fusarium oxysporum were classified into another clade,with the sequence similarity of 100%.And strains of F83,F97 and Fusarium incarnatum were classified into the third clade. The sequence similarity inside each clade were all higher,being 97.61%~100%.But the sequence similarity of strains among different clades were lower.[Conclusion] The phylogenetic analysis of ITS sequence could provide the important reference basis for the genus identification of Fusarium spp.
