Title: Effective dose for chemical Kinding model of epilepsy induced by bemegride in rats
Abstract: Objective To establish a kindling model of epilepsy induced by intraperitoneal injection of bemegride in rats and study its effective dosage. Methods Adult male Wistar rats were divided into 7 groups, and the doses of bemegride for estimating kindling epilepsy were scheduled according Krber's method. The intensity of behavioral seizures was evaluated by the use of the six_point scoring system, and EEG was recorded.Mean time of seizure beginning(SB),reaching the first stage V seizure (RFSvS) and reaching the kindling criterion (RKC) were observed. Results Effective dose of induced epilepsy (97% of animals) was drawn 18.74 mg/kg.The mean of SB and RKC were 213±100 seconds and 288±109 seconds respectively. The kindling rate was 86.7%. Conclusion In the experimental condition,it is optimal to select 18.7 mg/kg as the dosage of bemegride for establishing kindling model.
