Title: Study on in Vitro Culture of Hypocotyl of Black Sesame
Abstract: The hypocotyl of aspectic seedings of the black sesame(Sesamum indicum DC,cv.Jinzhi No.3)was used as the explants.The MS basic medium,supplemented with varous combinations of 6-BA(2.0 mg/L),IAA(0.1 mg/L,0.2 mg/L,0.3 mg/L,0.4 mg/L or 0.5 mg/L) and NAA(0.5 mg/L,1.0 mg/L,1.5 mg/L or 2.0 mg/L) were used as the callus induction and regeneration media.The 1/2MS basic medium,added with varous combinations of NAA(0.1 mg/L,0.3 mg/L,0.5 mg/L),NAA(0.1 mg/L) and IAA(0.1 mg/L) was used as the rooting media.We studied the callus induced from the hypocotyl and regenerated plants differentiated from the callus in the role of different media.The results showed that when inductive differentiation medium was MS+2.0 mg/L 6-BA+0.2 mg/L IAA,the average induction rate was the highest.Some hypocotyl can cut off the morphological top to form buds.When the rooting medium was 1/2 MS + 0.1 mg/L NAA,the rooting rate was 100%.
