Title: Molecular identification of witches'-broom disease on sweet cherry
Abstract: 【Objective】 Total DNA was extracted from main leaf veins of healthy and witches'-broom infected trees, to investigate if it was infected with phytoplasma or not. 【Method】 A nest PCR was processed using the special phytoplasma 16 S r RNA universal primer pairs P1/P7 and R16F2n/R16R2 followed. 【Result】 A 1.2 kb fragment was amplified from all the symptomatic plants, and no fragment was amplified from the asymptomatic plants. The PCR products were cloned and sequenced. The sequence was analyzed through the online tool i Phy Classifier and phylogenetic tree. Finally we found that the sweet cherry witches'-broom disease phytoplasma belongs to 16 Sr V-B subgroup, the same subgroup of Jujube witches'-broom phytoplasma, Peach yellows phytoplasma and Cherry lethal yellows phytoplasma. 【Conclusion】 This is the first report of 16 Sr V-B group phytoplasma associated with sweet cherry witches'-broom disease in China.
