Title: Effects of dioxins on aryl-hydrocarbon receptor and aryl-hydrocarbon receptor nuclear translocator expression in human sebaceous glands
Abstract: Objective To observe the effects of 2,3,7,8- tetrachlorodibenzo-p-dioxin(TCDD),a representative of dioxins family,on the expressions of aryl-hydrocarbon receptor and aromatic hydrocarbon receptor nuclear translocator(ARNT) protein in cultured human immortalized SZ95 sebocytes and human sebaceous glands. Methods SZ95 sebocytes were exposed to TCDD at the doses of 0(control) and 10 nmol/L respectively. After three-day incubation, immunocytochemistry method and Western blotting were used to study the effects of 10 nmol/L TCDD on AhR and ARNT proteins expression in SZ95 cells; Immunohistochemistry method was used to study the effects of 10 nmol/L TCDD on AhR and ARNT proteins expression in human facial sebaceous gland tissues. Results Compared with the control group,the intracellular AhR levels in SZ95 sebocytes treated by TCDD for three days were decreased and ARNT levels were enhanced significantly(P0.05). The expression of AhR decreased while the expression of ARNT was up-regulated after treatment with TCDD for three days. Conclusion Dioxin may induce the abnormal differentiation of sebaceous glands via AhR/ARNT signaling pathway.
