Title: The Effect on the Content of 5-HT and 5-HIAA in the Insomnia Rat′ Hypothalamus Caused by Sishencong(EX-HN1) Acupuncture
Abstract: Objective:To observe the content of 5-HT and 5-HIAA in the insomnia rats′ hypothalamus caused by Sishencong(EX-HN1) acupuncture,thus to reveal the central mechanism of the treatment for insomnia by acupuncture.Methods:Fifty male Wistar rats are divided into five groups at random:Blank control group(n=10);Model control group(n=10);Diazepam group(n=10);Hand acupuncture group(n=10);Electro-acupuncture group(n=10);Duplicate insomnia model of rats by means of PCPA abdominal injection.The animals in the two acupuncture groups will be treated with 7 days′ acupuncture,the animals in Diazepam group will be treated with seven days′ diazepam intragastric administration,then study the results of the treatments by observing the content of 5-HT,5-HIAA in the insomnia rats′ Hypothalamus.Results:After PCPA abdominal injection the normal sleep of rats disappeared,at the same time,the content of 5-HT and 5-HIAA in the hypothalamus of rats decreased with comparison to blank control group(P0.05).Model group compared with the blank,the content of 5-HT and 5-HIAA in the hypothalamus of rats decreased obviously,has significance difference(P0.01).There is no significant change between Diazepam group and Model control group(P0.05).Both hand acupuncture group and electro-acupuncture group can increase the content of 5-HT,5-HIAA in the insomnia rats′ hypothalamus,with significance difference comparison to model control group(P0.05),and have significance difference comparison to Diazepam group(P0.01).Compared with hand acupuncture group,electro-acupuncture group has significance difference(P0.05).Conclusion:Acupuncture can increase the content of 5-HT,5-HIAA in the insomnia rats′ hypothalamus.This may be one of the mechanisms that acupuncture can promote sleep and enhance the quality of it.
