Title: Molecular Detection of the Stem Rust Resistant Gene Sr33 in Spring Wheat Cultivars
Abstract: Sr33 is an important stem rust resistant gene,not only resistant to the domestic main stem rust races,but also for the new virulent pathogen Ug99(TTKSK) and its variant races TTKST and TTTKS.The aim of this study was to provide theoretical basis for the stem rust resistant gene Sr33 in wheat breeding programs.The microsatellite marker BARC152,which closely linked to Sr33 with good repeatability and high accuracy,was used to detect the presence of Sr33 gene in spring wheat germplasm and cultivars resistant to Ug99.The results indicated that only 1 genotype carried the resistant gene among the 58 improved Ug99-resistant spring wheat cultivars introduced from abroad,with a frequency of 1.7%,and 3 genotypes carried this resistant gene among the 18 main wheat cultivars in Heilongjiang province,with a frequency of 16.7%.It reflected that Sr33 was in a smaller proportion in introduced cultivars and main wheat cultivars in Heilongjiang,and much attention should be paid to the use of Sr33 in wheat breeding program.
