Title: Gene Expression Profiling Analysis of Mouse Liver Injury Caused by Qianliguang(Senecio scandens Buch.-Ham)
Abstract: OBJECTIVE To investigate the changes of the gene expression profile of mouse liver tissue by cDNA microarrays,and mechanisms of liver injury caused by Qianliguang at level of gene expression.METHODS After the mice were adminisered with Qianliguang extract at dose of 30 g·kg-1 per day for 15 and 30 d,respectively.The differentially,expressed genes significantly correlated with liver injury were screened and analyzed.The mechanisms of liver injury caused by Qianliguang were discussed especially at level of gene expression based on the biological functions of those differentially expressed genes and the relation between drug administration differentially expressed genes and physiological and biochemical index.According to the differentially expressed genes of different duration occurrence and development of liver injury induced by Qianliguang was analyzed.Furthermore,the key differentially expressed genes of different drug administration duration and their possibly physiological and pathological changes were also discussed.RESULTS 39 Differentially expressed genes found in the mice after taking 15-day Qianliguang.Among them,8 genes were up-regulated,while the other 31 genes were down-regulated.655 Differentially expressed genes were obtained in the mice after taking 30-day Qianliguang.Among them,337 genes were up-regulated while the other 318 genes were down-regulated.There were 21 common differentially expressed genes in both groups,including 2 genes up-regulated and 19 genes down-regulated.CONCLUSION The results indicated that the gene expression profiles of mouse liver cell could be changed significantly by Qianliguang,which might be important for elucidating the mechanisms of liver injury caused by Qianliguang.
