Abstract:Objective To observe the effects of γ-ray on MMP-9 mRNA and protein in RAW264.7 cells.Methods γ-ray(0,5,10,20 Gy) irradiated Mouse macrophage RAW264.7 cell line in vitro,which were divided into irradi...Objective To observe the effects of γ-ray on MMP-9 mRNA and protein in RAW264.7 cells.Methods γ-ray(0,5,10,20 Gy) irradiated Mouse macrophage RAW264.7 cell line in vitro,which were divided into irradiated and non-irradiated groups,6,12,24,48 hours after irradiation,Real-time polymerase chain reaction was used to detect the expression of MMP-9 mRNA,choose the dose-point with the most significant mRNA expression,6,12,24,48 hours after irradiation,Western blotting analysis was used to detect the expression of MMP-9.Dexamethasone(1 μM) was added to serum-free defined medium 1 hours before irradiation,Western blotting analysis was used to detect the expression of MMP-9 protein 24 hours after irradiation.Results Compared with control group,MMP-9 mRNA expression were significantly increased at 6,12,24,48 h post-irradiation(5,10,20 Gy)(P0.05),especially at 24 h post-irradiation with 10 Gy;MMP-9 protein expression were significantly increased at 6,24,48 h post-irradiation with 10 Gy(P0.05),especially at 24 h post-irradiation.Comparing with control group,dexamethasone could inhibit the MMP-9 protein expression which caused by irradiation.Conclusion Ionizing radiation can significantly increase the expression of MMP-9 mRNA and protein.Dexamethasone can effectively inhibit ionizing radiation-induced expression of MMP-9 protein.MMP-9 expression in RAW264.7 cells maybe involved in radiation-induced lung injury.Read More
