Title: Study of Radiosensitivity of Hepatocarcinoma Cell Lines in Vitro
Abstract: Objective To evaluate the radiosensitivity of hepatocarcinoma cells in vitro.Methods Using cell culture of human being hepatocarcinoma cell lines\| HepG2, SMMC\|7402 and SMMC\|7721, we measured their surviving fraction after being irradiated by 6MV X\|ray with doses from 0~10Gy by means of clonogenic assay and MTT. Radiosensitivity indexes have been calculated.ResultsThe D 0 of HepG2 is 1.5,SF 20.45±0.05Gy.The D 0 of 7402 is 1.6, SF 20.63±0.05Gy.The D 0 of 7721 is 1.8, SF 2 0.79±0.07Gy.There is a good correlation between the results derived from MTT and clonogenic assay( r=0.958,P0.05 ). Conclusion The radiosensitivity of HepG2 is rather high and 7402 and 7721 have lower radiosensitivity.MTT is a quick way to measure radiosensitivity of cells in vitro instead of clonogenic assay.
