Title: WT_1 mRNA Expression in Wilms Tumor by Fluorescence Quantity Reverse Transcriptase Polymerase Chain Reaction
Abstract: [Objective] To study the expression level of WT1 mRNA in Wilms tumor by fluorescence quantity reverse transcriptase polymerase chain reaction (FQ-RT-PCR) technique and its relationship with the bimolecular characteristic of WT and clinical features. [Methods] Twenty-two cases of WT (BWT 4, UWT 18, male 12 and female 10) were studied by quantitative determination of WT1 mRNA in tumor tissue, kidney and blood. Steps include: Total RNA study, cDNA synthesis,and FQ-RT-PCR. SPSS is used for statistics. [Result] WT1 mRNA has a higher quantitative expression level in WT tissue over kidney tissue or blood, UH WT over FH WT, blood of BWT over UWT, and Kidney tissue over blood. No difference of WT1 mRNA expression in blood was found between WT patients and normal children. [Conclusion] Expression level of WT1 mRNA in WT tissue play a role in the development of WT and is of relationship with WT pathological type and prognosis. It may become a prognostic factor, but has no relationship with WT patients' age, sex, BWT or UWT. Nevertheless, WT1 mRNA expression level in patients' blood may possibly be an indicator for BWT.
