Title: Effects of TSA on human hepatocellular carcinoma apoptosis and cells cycle
Abstract: OBJECTIVE:To investigate the effects of trichostatin A(TSA) on the apoptosis and cell cycle effect of human hepatocellular carcinoma(HCC) SMMC-7721 cells strain.METHODS:Light microscopic was used for the observation of cell morphology;Cell viability was determined by MTT assay;Flow cytometry(FCM) was used to investigate the inhibition and cell cycle;The expression of HDAC1 mRNA was detected by semi-quantitative RT-PCR.RESULTS:The multiplication speed of TSA treated cell was steped down and cell morphology was changed;MTT assay showed that TSA inhibited the proliferation of SMMC-7721 cells in dose-dependent and time-dependent ways;FCM analysis showed that the apoptotic rate increased in different concentrations of TSA reacted for 24 h and the early apoptotic rate was significantly increased from 8.72% to 18.22%(P0.026),and in the TSA treated group,G 0/G 1 phase cells significantly increased from 28.97% to 52.31%(P0.043),S phase cells increased from 26.01% to 34.28%(P0.051),and G 2/M phase cells decreased from 45.02% to 13.41%(P0.036).FCM analysis showed TSA could arrest SMMC-7721 cells at G 0/G 1 phase;RT-PCR detection revealed that the expression of HDAC1mRNA was down-regulated remarkably in the cells treated with TSA(from 0.34±0.02 to 0.11±0.01,P0.020).CONCLUSIONS:TSA can inhibit proliferation and induce apoptosis of HCC SMMC-7721 cells remarkably.It may be inhibiting enzymatic activity of HDAC1 and down-regulating the expression of HDAC1mRNA as well as arresting the cell cycles.
