Title: Inhibitory effect of EGCG combined with curcumin on proliferation of human ovarian cancer cells
Abstract: Objective To investigate the inhibitory effect and underlying mechanism of epigallocatechin gallate(EGCG)combined with curcumin on the proliferation of human ovarian cancer cell line A2780.Methods A2780 cells were randomly divided into four groups of A(blank control),B(treated with EGCG 10μmol/L),C(treated with curcumin 5μmol/L)and D(treated with EGCG10μmol/L plus curcumin 5μmol/L).The cell survival was measured by MTT assay,and DNA replication in A2780 cells was determined by EdU fluorescence staining.The single cell viability was observed by colony formation assay.The expressions of Cyclin B1 and Cyclin D1 were detected by Western blot.Results Both EGCG and curcumin could inhibit cell proliferation in a dose-dependent manner(P0.05).Compared with groups of A,B and C,the cell survival,positive cells of EdU staining,rate of cell colony formation and expressions of Cyclin B1 and Cyclin D1 were significantly decreased in group D(P0.05 or P0.01).Conclusion EGCG combined with curcumin can remarkably inhibit the proliferation of A2780 cells,which may be related with cell cycle arrest.
