Title: Effects of glucocorticoid on Muc5ac mRNA and protein expression in asthmatic mice lung
Abstract: Aim: To study the effects of glucocorticoid on the expression of Muc5ac mRNA and protein in asthmatic mice lung, and to investigate its role in mucus secretion of airways. Methods: Eighteen pathogen free BALB/c mice were randomly divided into control group, asthmatic group, and dexamethasone group. Dexamethasone group was treated with dexamethasone.The expression of Muc5ac mRNA and Muc5ac apomucin in lung were detected by the methods of reverse transcription polymerase chain reaction (RT PCR) and immunohistochemical assay, respectively. Results: The expression of Muc5ac mRNA(0.534 1±0.030 3)and protein (0.190 6±0.000 8) in asthmatic group were significantly higher than those of control group(0.199 4±0.012 8 and 0.119 4±0.000 7,respectively)and reduced(0.272 9±0.034 5 and 0.145 6 ±0.000 3,respectively)after treatment with dexamethasone. Conclusion: Dexamethasone substantially downregulates Muc5ac mRNA and protein expression, which may have potential therapeutic role in mucus overproduction of airways in asthma.
