Title: Effects of Simvastatin on the DNA Synthesis of Rat Cardiac Fibroblasts
Abstract: [Objective]To investigate the effects of simvastatin on the DNA synthesis of rat cardiac fibroblasts (CFs).[Methods]CFs of neonatal Sprague-Dawley (SD) rats were isolated by trypsin digestion method and growth-arrested CFs were stimulated with varying concentrations and time of Sim. DNA synthesis was measured by 3H-Thymidine (3H-TdR) incorporation.[Results]The results showed that: ①Sim decreased 3H-thymidine incorporation in cardiac fibroblasts in a concentration dependent manner.3H-TdR incorporation values of 10 -6 mol/L sim and 10 -5 mol/L sim group(1175±202.66 cpm/2000 cells,771±164.86 cpm/2000 cells,respectively) were significantly lower than that of the control (1608±204.32 cpm/2000cells) (both P 0.01 ); ②With the increase of time of the effects of 10 -5 mol/L Sim on CFs (6h, 12h, 24h, 36h, 48h), 3H-TdR incorporation of CFs decreased gradually, there was obviously negative relevance between 3H-TdR incorporation and time(r=-919,P0.01).[Conclusion]The results indicate that Sim decreases DNA synthesis and CFs number.
