Title: Protective effects of propofol on pulmonary ischemia-reperfusion injury in rats
Abstract: Objective To investigate the effects of propofol on ischemia-reperfusion(I-R)injury in rat lung and its mechanism. Methods A rat model of pulmonary warm I-R injury was established by occlusion of the left lung hilus for 45 min and then reperfused for 2 h.Animals were divided into four groups randomly:sham operation group(without occlusion),ischemia group(without reperfusion),I-R group, and propofol group(I-R with propofol anesthesia). We measured PaO2,wet to dry(W/D)weight ratio of left lung, the level of MDA,total protein and phospholipid in bronchoaveolar lavage fluid(BALF)of left lung, the CD18 expression and respiratory burst(RB)of neutrophils by flow cytometry. Results There were no significant differences in PaO2 among four groups. The levels of MDA,total protein in BALF and W/D ratio in I-R group were significantly higher than those in propofol group and sham operation group(P0.05).The level of phospholipid in BALF was lower than that in propofol group and sham operation group(P0.01).CD18 expression and RB of neutrophils in I-R group were higher than those in propofol group(P0.05).Conclusion Propofol had a protective effect on pulmonary I-R injury in rats. The mechanism may be related to the antioxidant effect and the suppression of the neutrophils activity.
