Abstract:Objective To observe the protective effect of ebselen on oxidative stress in endotoxin-induced acute lung injury.Methods Healthy male SD rats were randomly divided into six groups:normal control group...Objective To observe the protective effect of ebselen on oxidative stress in endotoxin-induced acute lung injury.Methods Healthy male SD rats were randomly divided into six groups:normal control group(C group),model group(LPS),ebselen low-dose group(EbsL group),middle dose group(EbsM group),high-dose group(EbsH group) and dexamethasone(DEX group).By intravenous injection of LPS(5 mg/kg) model,the treatment group in modeling the first 30 minutes,rats and ebselen(7.5 mg/kg,15 mg/kg,30 mg/kg),dexamethasone(5 mg/kg),normal control group and model group were injected into the same amount of solvent.6 hours after modeling,the animals were anesthetized and bleeding killed.The lung tissue were collected to detect W/D and the levels of SOD、MDA and MPO in lung homogenates and the change of light microscopy of lung histopathology.Results Ebselen treatment group compared to model group,the W/D were significantly reduced;the levels of SOD in lung tissue were significantly increased(P0.05);the levels of MDA and MPO in lung tissue were significantly decreased(P0.05);the pathology improved significantly.Conclusion Ebselen has a protective effect on endotoxin induced acute lung injury and its mechanism related with the inhibition of oxidative stress.Read More
