Title: Glucagon-like peptide-1 regulates the insulin gene expression in isolated rat islets
Abstract: Objective To investigate the effect of different concentrations of glucagon-like peptide-1(GLP-1) on insulin gene expression in isolated rat islets,and study the therapeutic roles of GLP-1 in type 2 diabetes mellitus.Methods GLP-1 was added at final concentrations of 1,10,10~2 and 10~3nmol/L to media containing 2.2mmol/L(low),5.5mmol/L(normal),11.1mmol/L(high) glucose.All islets samples were harvested after 24h,and total RNA was extracted in a single series for RT-PCR.Results At low and normal glucose concentrations,GLP-1 increased insulin mRNA(level in) a bell-shaped fashion.At high glucose concentrations,a dose-dependent increase in insulin(mRNA) levels was observed.Conclusion Insulin mRNA level is upregulated by GLP-1 dose-(dependently) and glucose-dependently.
