Methods Immunohistochemical staining techniques (ABC methods) and dou...Objective To study the immunohistochemical localization of Nogo-A in the spinal cord and dorsal root ganglion of normal adult rat. Methods Immunohistochemical staining techniques (ABC methods) and double staining immunofluorescence techniques were used. Results There is a widespread expression of Nogo-A in the oligodendrocyte, motor neurons and inner neurons in the grey matter of the spinal cord of adult rat. Positive structures mainly located in the cytoplasm and some processes. Nogo-A was highly expressed in the axons and the myelin sheath surrounding fibers which pass through the white matter of spinal cord. Also Nogo-A immunoreactivity was intense in myelinated and unmyelinated axons in the ventral root, dorsal root and sciatic nerve. Similarly, most of dorsal root ganglion neurons expressed Nogo-A, with different immunoreactivity levels. The positive structures present in the cytoplasm and neurites of large, medium and small sense neurons. Conclusion The widespread expression of Nogo-A in spinal cord, dorsal root ganglion and peripheral nerves indicates that it possibly plays an important role on the functional activity of the nerve system in the normal situation.Read More
