Methods: Hela cells were used as in vitro models. t...Objective: To understand if apoptosis induced by arsenic trioxide(As2O3) is related to the alteration in mitochondrial tranmembrane pitentials(ΔΨm). Methods: Hela cells were used as in vitro models. theΔΨm was detected by the double staining of propidium iodide(PI) and Rhadamine 123 (Rh123), and apoptosis was confirmed by cell viability,staining of hematomylin-eosin(HE), flow cytometry as well as gel electrophoresis of genomic DNA and morphological observation. Results: After treatment of Hela cells with As2O3, cell viability observed by trypan-blue exclusion was decreased, and the number of apoptosis cells measured by flow cytometry was increased apparently, and the morphotogical characteristic of apoptosis was detected by the staining of hematomylin-eosin(HE), and DNA of treated Hela cells showed a ladder parrern and the ΔΨm was disrupted with As2O3 treatment(P0.01). Conclusion: the mechanism of apoptosis induced by As2O3 may be related to the loss of ΔΨm.Read More
