Title: Regulation of MDI301 on the expression of MMP-1,MMP-2,TIMP-1 and procollagen in human fibroblast cells
Abstract: Objective: To detect the expression of matrix metalloproteinase-1(MMP-1),matrix metalloproteinase-2(MMP-2),tissue inhibitor of metalloproteinase-1(TIMP-1) and procollagen type I and III in normal and high glucose cells when treated with MDI301.And explore the possible effect of MDI301 in diabetic skin and foot ulcers.Methods: The gene expression levels of MMP-1,MMP-2 and TIMP-1 were detected by Real time PCR;The TIMP-1 production in human fibroblast cell cultured media was measured by ELISA kit;Procollagen type I and III protein levels were detected by Western blotting.Results: The mRNA levels of MMP-1 and MMP-2 in high glucose cells could be reduced when treated with MDI301(P0.05);The mRNA and protein expression of TIMP-1 and Type I,III procollagen in high glucose cells were increased with the treatment of MDI301(P0.05).Conclusion: MDI301 may have similar effect on the influence of MMP-1,MMP-2,TIMP-1,procollagen type I and III in diabetic patients,which may be used for the treatment of diabetic skin and foot ulcers.
