Title: Effect of octreotide on proliferation of human hepatocellular carcinoma SMMC-7721 in vitro and relevant mechanisms
Abstract: Objective To study the effect of somatostatin analogue octreotide on proliferation of human hepatocellular carcinoma SMMC 7721 in vitro and to study the relevant mechanisms on the cell cycle regulation.Methods The MTT was used to estimate the killing effect of octreotide on proliferation.The cell cycle distribution was analyzed by flow cytometry(FCM).The immunohistochemistry was used to detected protein expression of cyclin A、PCNA.Results The growth of SMMC 7721 was significantly inhibited in a marked dose dependent manner;Compared with control group,octreotide could cause the spano mumber in G 0/G 1、G 2/M phase to decrease and the auxo number in S phase of experiment group increase.After the cells were incubated with octreotide for 72h, the level of expressions of cyclineA、PCNA protein was decreased significantly(control:2 833±0 074,4 851±0 059;octreotide:1 67±0 043,3 257±0 036).Conclusion Octreotide can disturb cell cycle progression and inhibit cell proliferation through down regulation of the expression of cycline A、PCNA.
