Title: Study on the Analytical Methods of Water Soluble Sugars in Tobacco
Abstract: [Objective] The research aimed to study the influencing factors of water soluble sugar determination in tobacco,set up the determination methods of water soluble sugar in tobacco and provide the theoretical support for judging the sensory quality of cigarette.[Method]1.000 0 g tobacco samples were transferred into a taper flask containing purity water,after ultrasonic extraction 30 min at 40 ℃ and filter,then the filtrate were analyzed by liquid chromatography with evaporative light scattering detector(HPLC-ELSD).The effects of extraction time,extraction temperature,mobile phase,ELSD parameter on the determined value of water soluble sugar were studied.[Result] The experimental results showed that ultrasonic extraction time was 30 min,temperature was 40 ℃,acetonitrile-water was used as the mobile phase and the column temperature was 30 ℃,the drift tube temperature of ELSD was 80 ℃ and the air flow rate was 2.00 L/min.The proposed HPLC-ELSD method had a good linearity from 0.2-20.0 μg of fructose,flucose,sucrose and maltose in the leaching solution,with the detection limits of 12,10,14,20 ng respectively.The relative standard deviation of the method for each kind of sugar was less than 5%.[Conclusion] The method is accurate,rapid and has been applied for the quantitative analysis of soluble carbohydrates in tobacco.
