Title: Effects of tetrandrine on apoptosis and Caspase-3 protein expression in mouse lung fibroblast
Abstract: Objective To explore the effect of tetrandrine( Tet) on the apoptosis expression of and Caspase-3 protein in mouse lung fibroblast cells( L929). Methods Lung fibroblast cells( L929) lines were incubated in 50 m L · L~(-1)CO_2 cells incubtion box at 37 ℃ for passage culture. Fibroblast cells were divided into four group: blank group and different does Tet( 2. 5,5. 0,7. 5 mg · L~(-1)) groups which were cultured in Roswell Park Memorial Institute1640 medium without fetal bovine serum,penicilin and streptomycin. The cells were collected after 24 h,then morphological changes of the cells were observed under the transmission electron microscope. The cell apoptosis was detected by the flow cytometry.And the expression levels of Caspase-3 protein were examined with immunohistochemistry method. Results The apoptotic cells increased while the number of cells decreased in different Tet concentrationgs group,when compared with the blank group. There were significant different in the apoptotic cells compared with blank groups( P 0. 05). The apoptosis rate of blank group were( 5. 32 ± 0. 50) %,different does of Tet were( 7. 00 ± 0. 48) %,( 9. 66 ± 0. 76) %,( 20. 36 ± 1. 11) %. There was significant different in the Caspase-3 expression among the differentgroups( P 0. 05),and as Tet dosage increased,the expression of Caspase-3 protein increased( P 0. 05).Conclusion Tet can promote the apoptosis of mouse lung fibroblast cells and increase the expression of Caspase-3protein so that the lung fibroblast was inhibited.
