Title: Study on the Seed Germination of Swertia mileensis
Abstract: [Objective] The study was to provide the technical basis for preserving and artificial domesticating and cultivating Swertia mileensis seeds.[Method] The seeds of S.mileensis were collected from Minle mountain in Yunnan,and its 1 000-seed weight was measured after natural dry for 20 d.The number of germinating seeds were recorded after being cultivated for 30 d under the conditions of different temperature(15,20,25,30,35,40 ℃) and different illumination(red,yellow,blue,green,white and dark).The seed germination rate was calculated after the seeds being soaked for 3.0 h in GA3(100,200 mg/L GA3 solution),3% KNO3 solution and distilled water(CK) and being cultivated for 30 d under(25±1) ℃ and natural light.[Result] The suitable temperature for germination of S.mileensis seeds was 20-30 ℃.The illumination had promoting effect on seed germination.The seed germination rate was the highest after being stored for 4 months under the condition of natural state.The seed longevity was about 7 months,and the seed longevity could be prolonged by low temperature under 4 ℃.The seed dormancy could be broken,and the seed germination rate could be increased significantly with the method of seed soaking with GA3 and KNO3.[Conclusion] The results of the test were consistent with that of most of related studies.
