Title: Biological Characteristics of Human Bone Marrow-derived Mesenchymal Stem Cells
Abstract: Objective To establish a method of isolation and cultivation of human bone marrow mesenchymal stem cells(MSCs)in vitro and investigate the biological characteristics of MSCs. Methods Human fetal MSCs were isolated and cultured in DMEM. The morphology of MSCs was observed with inserted microscope constantly. Cell cycle and surface marker were analyzed by flow cytometry. The growth characteristic of MSCs was measured during primary and passage culture. Results MSCs showed active proliferative ability in primary and passage cultere before the 14 th . Cell cycle analysis showed that 82% of MSCs was in G 0/G 1 phase. The cells were positive for CD 44 and negative for CD 34 ,CD 45 . Conclusion Before the 14 th passages,MSCs showed stable and rapid growth in vitro, MSCs could be used as the seed cells for tisuue engineering.
