Title: Induction of apoptosis with TRAIL in Non-small cell lung cancer
Abstract: Objective To investigate whether TNF-related apoptosis-inducing ligand (TRAIL) can induce apoptosis in Non-small cell lung cancer cells. Methods NSCLC cell line NC1-H460 was treated with TRAIL in serial concentrations from 20 ng/ml to 160 ng/ml. Caspase 3 activity was measured after 8 hours of the treatment. Morphological changes of the nuclei were investigated using Fluorescence microscope and the sub Gl fraction of the cells was determined with Flow cytometry in 24 hours. Results TRAIL resulted in a dose-dependent increase in Caspase 3 activity in eight hours. Karyopyknosis and karyorrhexis were noticed in 24 hours in the TRAIL treated cells. These morphological changes as well as the sub Gl fraction of cells were more distinctive with the increase of TRAIL concentration. Conclusion The data suggest that TRAIL is a potent inducer of apoptosis in the NSCLC cell line NC1-H460 in vitro.
