Abstract:Objective:To investigate the cytotoxicity of lidocaine to SH-SY5Y cells by detecting release rate of lactate dehydrogenase(LDH),apoptosis rate and caspase-3 expression level.Methods: SH-SY5Y cells wer...Objective:To investigate the cytotoxicity of lidocaine to SH-SY5Y cells by detecting release rate of lactate dehydrogenase(LDH),apoptosis rate and caspase-3 expression level.Methods: SH-SY5Y cells were treated with 0%,0.5%,1% and 2% lidocaine for 10 min.Release rate of LDH,apoptosis rate and caspase-3 expression level were detected.Results: Release rate of LDH was 11.7% in control cells.LDH release rate was significantly increased up to 18.8% and 28.5%,respectively,after treatment with 1% and 2% lidocaine.Apoptosis rate was 6.1%,14.9%,19.2 % and 23.0%,respectively,after treatment with 0%,0.5%,1% and 2% lidocaine.Expression of caspase-3 in control group was lower than that in lidocaine groups.Conclusion: Lidocaine causes SH-SY5Y cell injury at concentrations of 0.5%,1% and 2%,and the cell injury becomes more obvious with increasing of the concentration.Read More
