Title: Effect of different doses of sensibiligen on childhood rats of atopic asthma
Abstract: 【Objective】 Sensitize the Wistar Rats in childhood with ovalbumin,and set a rat model in childhood of atopic asthma,to investigate the effect of different doses of sensitinogen on airway allergic inflammation in the childhood rat model.【Methods】 The asthma model of Wistar rats in childhood were prepared by ovalbumin(OVA) and alminum hydroxide(Alum) with a low,a middle and a high dose respectively.The pathologic changes of lung tissue,the total cell count in bronchoalveolar lavage fluid(BALF) were observed.The concentrations of IL-4 in serum and OVA-IgE in BALF were measured by using sandwich ELISA.【Results】 Low-dose,middle-dose and high-dose,the three sensitized model groups suffered from asthma abnormal symptoms compared with control group,and the high dose group had a significant abdominal breathing and shortness of breath.The inflammation in high dose group was more severe than both low dose group and middle dose group.Total cell count of BALF in the three sensitized groups were increased,to varying degree,compared with control group,most significantly in high dose group(P 0.05).The serum IL-4 and BALF OVA-specific IgE were augmented significantly in high dose group compared with control group,low dose group and middle dose group(P 0.05),and the BALF OVA-specific IgE was higher than control group as well(P 0.05).【Conclusion】 The doses of antigen in a childhood rat model of atopic asthma are related with the extent of airway allergic inflammation.The more antigen it is sensitized,the severer airway allergic inflammation is.
