Title: Study on the Browning Phenomenon of Walnut Explants in Vitro Culture
Abstract: Study was carried out on the factors resulting in browning of walnut explants in vitro culture. The result showed that new shoots collected in April have severer browning and a higher death rate while the shoots collected in June are more effective for culture, with the dormant buds to be second to them.In vitro culture, the browning of explants could be reduced with the liquid medium. With the help of hor- mones added at proper concentrations (6-BA 1.0mg/L,IAA 0.2mg/L or GA 0.5mg/L) explants can have a better germination of auxiliary buds, with the browning phenomena lessened.Addition of Na2S203 to medium can effectively prevent walnut explants from browning, and promote germination and shoot growth.
