Title: Diversity of Cellulose-producing Thermophilic Bacteria in the Baoshan Hot Spring
Abstract: To obtain amylase-producing,cellulase-producing and protease-producing thermophilic bacteria,two kinds of separation medium were used on different temperature and pH by spread-pate technique.16S rRNA genes of these bacteria were obtained by PCR amplification with universal primers.The obtained PCR products were sequenced and blasted in GenBank.Based on 16S rRNA gene sequences,3 phylogenetic trees were constructed to analyze the taxonomy of thermophilic bacteria.All together 200 cellulose-producing strains were isolated from the two main hot springs in Baoshan,Yunnan Province of China.All the 200 strains belonged to 30 species.Among them 122 stains were isolated from culture conditions of 55 ℃,78 stains from culture conditions of 65 ℃,belonging to 15 species.The result showed that more than 95% cellulase-producing strains produce amylase,and about 50% of cellulase-producing strains produce protease.In hot spring,cellulase-producing,amylase-producing,and protease-producing strains exist extensively.
