Title: Screening Study on Penetration Enhancers of Kechuanxuewei Patch
Abstract: OBJECTIVE:To explore the optimal concentration of the penetration enhancers-laurocapam(Azone)for Kechuanxuewei patch, and to screen the best technological conditions.METHODS:Using the cumulative permeation percentage of Eugend as assessment parameter,an in vitro transdermal test was performed on the extracorporeal rabbit skin in a special improved Franz(one-compartment)diffusion cell to identify the penetration speed and permeation of Azone at the amounts of 0%,1% and 3%.A transdermal dynamics model was established to extract skin penetration parameters and compare the indexes.RESULTS:The dynamics model of penetration rabbit skin rate meets Higuchi equation through mathematic disposal treatment and statistical analysis, when the amounts of Azone were at 0% and 1% ;the model was in line with the Weibull distribution model when the amount of Azone was at 3%.When 1% Azone was added, the test showed the fastest speed in skin penetration,5.019 times that without Azone, and generated the most penetration concentration,2.489 times that without Azone.CONCLUSION:When 1% Azone was added, the test showed the fastest speed in skin penetration and generated the most penetration concentration.
