Methods:Orthotopic liver transplantation i...Objective:To study and reform of orthotopic liver transplantation in rat, and to observe allograft rejection of liver transplantation in Wistar rat operated. Methods:Orthotopic liver transplantation in 30 Wistar rats which received SD liver were established by using the two-cuff technique with some modifications in cuff tube, recipients anaesthesia and anastomoses of suprahepatic vena cava, portal vein, infrahepatic vena cava. The thirty rats after transplantation were randomly divided into control (n=5) and therapy group (n=15), and latter was subcutaneously injected by cyclosporine 2 mg/kg daily for 1 week after operation. In meantime, allografts rejection of all transplantation rats were studies. Results:The time of vascular anastomosis for suprahepatic vena cava was 6.5 to 10 minutes, anhepatic period was 8.5 to 12 minutes, more than 93% of grafted animals survived for seven days. The control group all died 9 to 13 days after transplantation, and their histopathologic examination showed typical allografts rejection. Up to now, cyclosporine therapy group are surviving. Conclusion:The modified can increase survival rate of liver transplantation in rat. The donor liver of SD rats and recipient Wistar rats combination is a high rejection model. It can be effectively used to study allograft rejection and liver transplantation tolerance.Read More
