Title: COMPARISON STUDY OF GENOME DNA EXTRACTION METHODS FROM CYANOBACTERIUM
Abstract: With the widespread use of molecular techniques in microalgal community,one of important issues is DNA extraction and preservation.In this paper,three DNA extraction methods(the representative commercial kit,the CTAB,and the Wizard methods)applied to the freshwater cyanobacterium Microcystis viridis were compared based on the extraction efficiency,purity,quantity,and integrity of DNA extracts,as well as amplify-ability.The results indicated that the most efficient,economic,and convenient DNA extraction method is CTAB,although both CTAB and Wizard methods produced high yield and purity of DNA suitable for PCR amplification,cloning,and enzymolysis.On the other hand,we found that alcohol fixation for microcystis cells is a good preservation strategy.By the optimization of fixation concentration,fixation time,and fixation temperature,high quality of DNA can be obtained to be used as the source of nucleic acids for further molecular analyses.
