Title: Analysis of the results of antinuclear antibody and anti-ENA antibodies for 979 patients of autoimmune diseases with IIF and biochip technology
Abstract: Objective To analyze the relationship between antinuclear antibody(ANA) and anti-extractable nuclear antigens(ENA) antibodies in autoimmune diseases. Methods ANA and anti-ENA(ds-DNA、Sm、SS-A/Ro、SS-B/La、u1RNP、Scl-70、Jo-1、Ribosomal P)antibodies were detected by indirect immunofluorescence(IIF) and biochip technology, respectively. Results Of all the 979 samples,400 cases were positive for ANA,the positive rate was 40.9%(400/979).The majority of patterns were speckled pattern(249 cases,62.25%),homogeneous pattern(83 cases,20.75%),and cytoplasmic granular pattern(30 cases,7.5%),respectively.Anti-ENA antibodies were detected in 979 cases simultaneously, and 199 cases were positive. The positive rate was 20.3%(199/979),and only one case is negative for ANA.Of the 400 cases, the positive rate of the anti-ENA antibodies was 49.5%.The most frequently occurred antibodies were anti-SS-A/Ro(133 cases),anti-Rib P(78 cases),anti-u1RNP(70 cases),anti-ds-DNA(61 cases)and anti-SS-B/La(40 cases) antibodies, mainly associated with the speckled pattern. In the cases of homogeneous pattern for ANA,the common anti-ENA antibodies were anti-ds-DNA(37 cases),anti-SS-A/Ro(27 cases),anti-Jo-1(20 cases) and anti-Rib P(20 cases) antibodies. Conclusion In order to accurately determine the target antigens,it is necessary to screen ANA with IIF and to confirm the types by anti-ENA antibodies.
