Title: The Construction of cDNA PCR Library from a Small Amount of RNA
Abstract: By the method of PCR (Polymerase Chain Reaction), we have constructed the cDNA PCR library from mRNA. The cDNA PCR library can amplify the original cDNA up to hundreds of times. With the total RNA of human K562 cultured cell, the cDNA of beta-Actin has been obtained by the methods of cDNA PCR library and reverse transcription respectively. As contrast, the amount of beta-Actin^s cDNA from the cDNA PCR library is much higher than from reverse transcription. 75pg total RNA of human K452 Cultured cell is employed to construct 50mul cDNA PCR library, and the cDNA of beta-Actin can even be detected by using 1mul of the library as template to perform the PCR. Therefore cDNA PCR library can greatly enlarge the amount of information.
