Title: Effect of resveratrol on apoptosis of lung adenocarcinoma cell line A549
Abstract: OBJECTIVE:To investigate the function of resveratrol(Res)and its mechanism in lung adenocarcinoma A549cells.METHODS:A549cells were treated with resveratrol at different concentretions for 24hand the morphological change was observed by phase contrast microscope.Apoptosis features were observed by fluorescence microscope after nuclear DAPI staining.The proliferation of A549cells was analyzed using methyl thiazolyl tetrazolium(MTT)assay.The expression of p53,Bax,Bcl-2and Cleaved-Caspase3after Res treatment in A549cells was measured by Western blot.RESULTS:After Res treatment,we found that the interspace of A549cells was decreased and granular material was found in nucleus.These changes were remarkably associated with the increase of resveratrol concentration.DAPI staining indicated that Res induces apoptosis in A549cells.After treated with 25-200μmol/L Res,the proliferation was markedly inhibited with a does dependent manner(25μmol/L,12.4%;50μmol/L,21.6%;100μmol/L,51.3%;200μmol/L,89.7%),and IC50 was 98μmol/L.Otherwise,after Res treatment,the expression of p53,Bax and Cleaved-Caspase3protein was upregulated(r=0.930,P0.001;r=0.874,P0.001),Bcl-2and the ratio of Bcl-2/Bax were downregulated(r=0.753,P= 0.01).CONCLUSION:Resveratrol could inhibit the proliferation and induce apoptosis through p53method in A549cells, and the expression of Bax,Bcl-2and Cleaved-Caspase3participate in the apoptotic process modulated by resveratrol.
