Abstract: An in vitro protocol was developed for multiplication of Dioscorea rotundata by using leaves,stem,tuber and adventitious root derived from plants grown in pot as explants.Effects of different plant growth regulators on callus induction,callus differentiation,adventitious bud induction and axillary bud multiplication were studied.The results showed that the optimum medium for proliferation of axillary bud located below the shoot tip was MS+BA 3.0mg/L+NAA 0.2mg/L(3.1 for coefficient of multiplication).The optimum medium for callus induction was MS+2,4-D 4mg/L+6-BA 0.5mg/L,and the rate of callus inducement was as high as 85.7% when stems with axillary buds were used as explants.When the concentration of BA was 4.0~6.0mg/L,the rate of adventitious bud formation could up to 97.5%.Increasing the concentration of BA and agar could decrease browning during the culture.
