Title: Effects of Kechuan Liuwei Mixture on Expression of IL-17A mRNA and Airway Inflammation of Asthmatic Mice Induced by RSV
Abstract: Objective:To investigate the role of Kechuan Liuwei Mixture in the development of RSV-induced,OVA-sensitized airway inflammation and the expression of IL-17A mRNA in lung,so as to explore its underlying mechanism.Methods:40 mice were randomly divided into four groups:PBS group,OVA group,OVA/RSV group and KLH group.The asthma model was reproduced by sensitization with intraperitoneal injection of OVA,followed by repeated inhalation of OVA combined with repeated intranasal instillation of RSV(1.0×106 PFU/mL in 50 μL).They were detected by RT-PCR which were interferon gamma(IFN-γ),interleukin-4(IL-4) and interleukin-17A(IL-17A) mRNA in lung of mice.The changes of the lung histopathology were evaluated by HE stain.Results:In comparison with PBS group,IFN-γ was decreased significantly,IL-13 and IL-17A were increased significantly in OVA group(P0.05,P0.01),and IL-13 and IL-17A were also increased significantly in OVA/RSV group(P0.05,P0.01),but there was no significant difference in IFN-γ.In comparison with OVA/RSV group,IFN-γ was significantly increased and IL-13,IL-17A were decreased significantly in KLH group(P0.05,P0.01).Conclusion:Kechuan Liuwei Mixture can control the inflammation of the airway,and regulate the expression of cytokines in asthmatic mice.
