Title: Effects of rhG-CSF on HIF-1a expression in hypoxia ischemia brain damage in neonatal rats
Abstract: Objective:To investigate the effects of rhG-CSF on the HIF-1a expression in hypoxia ischemia brain damage(HIBD) in neonatal rats.Methods:The Wistar rats,aged 7 days,were divided into Sham-operation group,HIBD group and rhG-CSF group,respectively.The rats of the HIBD group were anesthetized with ethylether.The right common carotid artery was exposed and ligated.Then,they were exposed to hypoxia in a normobaric chamber for 2.5h.The HIF-1αprotein expression was detected by immunohistochemistry.Results:The HIF-1a expression of the HIBD group was increased compared with that of the sham-operation group(P0.01).The HIF-1αexpression of the rhG-CSF group was obviously increased compared with that of the HIBD group(P0.01).Conclusion:The up-regulation expression of HIF-1α may be involved in suppressing apoptosis,and rhG-CSF may enhance the tolerance of the brain tissue to hypoxia and reduce apoptosis possibly by regulating HIF-1αexpression level,suggesting that rhG-CSF can improve prognosis of HIBD rats.
